Information on Higenamine
Higenamine hcl, commonly known as demethyl coclaurine, and norcoclaurine in the chemical field. Higenamine is defined as a chemical compound found in a variety of plants including Nandina domestica, Aconitum carmichaelii, Asarum heterotropioides, Galium divaricatum, Annona squamosa, and Nelumbo nucifera (lotus seeds). Higenamine has a long history of use in traditional medicine in ancient China. Historically, the plants that higenamine comes from have been used to treat breathing disorders and cough through these receptors.
Higenamine, with its catecholamine-like structure, has been isolated in several plant species including including Nandia domestica (fruit), Aconitum carmichaeli or Aconitum japonicum (Aconite root), Asarum heterotropoides (root), Gnetum parvifolium (stem/vine), Anona squamosa (stem and leaves) and Nelumbo nucifera (seed).
Through the Î²-adrenoceptors higenamine mediates cardiotonic, vascular relaxation and bronchodilator effects. This compound particularly shows promise as a beta-2 adrenergic receptor agonist similar to that of ephedra or synepherine and has demonstrated the ability to increase the expression of cyclic adenosine monophosphate. This may result in a lipolytic effect from human consumption.

The murine group pretreated with higenamine, revealed a relaxation reaction of corpus cavernosum induced by the PDE-5 inhibitor increased significantly, compared with CC exposed to the PDE-5 inhibitor but not pretreated with higenamine.
Higenamine has also shown anti-platelet and anti-thrombotic effects. In the investigation done by the Seoul National University, an acute thrombotic condition was induced in mice by the injection of the mixture of collagen and epinephrine. Higenamine showed inhibitory activities to both human and rat platelet aggregation induced by ADP, collagen and epinephrine. The oral administration of higenamine had increased the recovery rates from the acute thrombotic challenge in mice.
In terms of toxicity, the compound is estimated to be present at approximately 47.5 mg in a 9 ounce serving of lotus (Nelumbo nucifera) seeds so it is estimated that the level of consumption occurring in food has already approximated that which will be used in the dietary supplement. However, in addition to this calculation, an acute toxicity study in mice revealed no deaths at a dose of 2 g/kg, indicating that the compound does not possess overt toxicity in an acute setting after oral administration.
